Development of hybrid P450 enzymes as light activated biocatalysts

Hybrid P450 Enzyme

Our long-term goal is to engineer hybrid enzymes capable of performing light-activated selective oxidation of organic substrates. We have developed hybrid enzymes by covalently attaching a Ru(II) photosensitizer to non-native single cysteine residues strategically positioned in close proximity to P450 BM3 heme domain mutants. As proof of concept, a series of hybrid enzymes has been shown to catalyze the hydroxylation of lauric acid upon light activation.
