Development of hybrid P450 enzymes as light activated biocatalysts.

Our long-term goal is to engineer hybrid P450 enzymes capable of performing light-activated selective oxidation of organic substrates. We developed hybrid enzymes by covalently attaching a light-harvesting unit, i.e. Ru(II) photosensitizer, to non-native single cysteine residues strategically positioned in close proximity to P450 BM3 heme domain mutants. A combination of rational design and directed evolution approaches are employed to engineer the next generation of hybrid P450 enzymes.
